The aim of this work was to obtain new medical biomaterials based on a biocompatible polymer polytetrafluoroethylene (PTFE), modified with the drug, ketoprofen (KP), and gold nanoparticles (Au NPs). The biological inertness of PTFE, hydrophobicity, elasticity, low weight prevents sticking to wounds and makes fibrous materials based on it ideal for dressing and obturators. Ketoprofen gives anti-inflammatory properties to the material, while Au NPs provides it with antibacterial ones.
Biomaterials based on porous PTFE containing Au NPs and KP were prepared using two green technologies, fluid technology and metal vapor synthesis (MVS). Porous fluoropolymer was synthesized in vacuum under CO2 laser radiation of PTFE. Fluid technology using supercritical carbon dioxide (SC CO2) was applied to form the nanoporous surface of the polymer and impregnate ketoprofen. Nano- and micropores were formed on the surface of the fluoropolymer by the micro-discontinuity method during decompression of SC CO2. The directional formation of porosity in PTFE was used to stabilize Au NPs deposited by MVS in the surface layer of the polymer.
MVS is an efficient means of purposeful synthesis of metal nanoparticles and composites containing them. The modification of polymers with metal nanoparticles gives rise to several attractive functional properties such as magnetic, catalytical and antibacterial. The nanostructured polymer was modified with the organosol of Au NPs in isopropanol prepared by MVS. Ketoprofen was introduced into the Au/PTFE system from a solution of SC CO2. Transmission electron microscopy (TEM) shows that the particles are of predominantly spherical shape, although the size distribution is relatively broad. The average size of Au NPs in Au/PTFE composite is 8.0 nm. The chemical state of Au atoms in the composites was determined by X-ray photoelectron spectroscopy (XPS). It has been established that Au NPs mainly stabilize on ketoprofen. This follows from the binding energy of the Au 4f7/2 peak which is 0.8 eV higher than that of Au foil. This shift could be attributed to both the size effect and the presence of the Au?O bond. However, the first hypothesis should be excluded because of the particle size.
The combination of SC CO2 and MVS with the involvement of laser radiation to produce metal-containing nanomaterials based on fluoropolymers impregnated with a medical anti-inflammatory drug was proposed for the first time.
