In the last years, the cultivation of methane-oxidizing organisms has been sort of great interest, because it allows to obtain value products using accessible raw materials, such as natural gas. The products obtained during the cultivation of mixed culture based on Methylococcus capsulatus can be used in agriculture, medicine and other fields. Nucleic acids and their derivatives are one of the most popular components that can be produced during the comprehensive processing of bacterial biomass. As a result of the extraction of nucleic acids from bacterial biomass, two valuable products are formed: nucleic acids itself and denuclearized biomass. The extraction of nucleic acids is carried out basically by salt and alkaline solutions. The extraction process conditions have a great impact on the degree of nucleic acids release from the biomass. Therefore, it is necessary to find optimal conditions of extraction to develop an effective process of obtaining useful products. We found out that the alkaline extraction of nucleic acids from the biomass of a methane-oxidizing microorganism consortium is more productive than the extraction by water solution of diammonium phosphate. However, in the case of alkaline extraction there is higher concentration of protein in the extract. As a result, optimal extraction conditions of nucleic acids by diammonium phosphate and sodium hydroxide solutions were selected. The article shows that the best extraction results are achieved when we use lyophilized biomass at temperature of 95 degrees Celsius, pH range of 8.5-9.0 and biomass concentration of 10%. The necessity of two-stage extraction for separation of DNA and RNA separately from each other is shown. The conditions for concentration process of extracts by ultrafiltration after the first and second stages of extraction were selected. The article also provides a scheme of obtaining denuclearized bacterial biomass and technical preparations of DNA and RNA.
